DR signal is the cable. The CNC end of the cable needs to have pins 6,8 and 20 all linked together. See the diagrams at
The message about port not available. If you have multiple DNC windows open then maybe one of the others is using com16?
How many com ports do you have? Close easyDnc and restart it. Then without opening any DNC windows go into easydnc setup and click the "detect ports" buttons and that will show a list of available ports and whether they're free or busy.

We do have pins linked we used a gender changer Null Adapter to link. If I reboot the machine and computer it will works for a few then gives me the same error.

As Bilbo said, How many com ports do you have? Close easyDnc and restart it. Then without opening any DNC windows go into easydnc setup and click the "detect ports" buttons and that will show a list of available ports and whether they're free or busy. What does it say.
And why com16? Are you using a multipprt serial adapter/box? Serial port server? windows terminal server? Remote desktop etc? basically what hardware is it?

Not sure what "G&I" is. But I'd say that at the end of the file the CNC is expecting some special 'end' character. In its simplest form used by most CNC's this is just a % percent symbol at the very end of the file. If your file doesn't have one try entering one before sending the file to the CNC.
Failing that you need to look through your documentation for the control. It will usually show the expected file (tape) format. When you know what special character you want you can enter the ASCII value for that character into the advanced settings of the easyDnc setup and easyDnc will insert that character automatically at the end of each upload to the CNC.
If you have the 'character' but don't know its ascii value come back here and tell us.
